ChapterDx MLSTnext Listeria genotyping NGS assay analyzes 69 most polymorphic loci spanning L. monocytogenes genome. The combination of polymorphic information at those loci provides high-resolution genotype for L. monocytogenes, enables detecting co-presence of different L. monocytogenes in the same sample. The assay can detect other Listeria species: L. grayi, L. innocua, L. ivanovii, L. marthii, L. seeligeri, and L. welshimeri

The total workflow is designed for efficiency, requiring less than 13 hours to complete. This timeline includes 4-5 hours of benchwork and incubation time, approximately 7 hours of sequencing, and less than 30 minutes for automated data analysis.

Technology

The ChapterDx MLSTnext™ Listeria Genotyping NGS Assay follows a streamlined workflow consisting of the following steps:

  1. DNA extraction (not provided)

  2. Multiplex PCR

  3. Pooling and purification

  4. Next-generation sequencing​

During the multiplex PCR reaction, target amplification and specimen indexing occur simultaneously within a single tube. Following the PCR reaction, amplicons from different samples are pooled together into a single tube for downstream processing. All subsequent procedures are performed on this single pooled tube, regardless of the initial number of samples.

Once the purification cleanup is complete, the pooled library is sequenced on an Illumina platform. The total workflow is designed for efficiency, requiring approximately less than 13 hours to complete.
