Chapter Diagnostics PomePlex Target Enrichment NGS assay is designed for high sensitivity using a dual-amplification strategy. cDNA samples are first amplified, then subjected to probe-based capture (Target Enrichment) using streptavidin beads. A second PCR step incorporates sequencing indexes. This method is suitable for small to high-throughput sample scales.

The assay procedure consists of the following steps: nucleic acid extraction (reagent not provided), reverse transcription, cDNA amplification, cleanup, target enrichment, amplification, cleanup, multiple sample pooling, and sequencing (reagent not provided). The turnaround time for the assay is less than 14 hours (6 hours laboratory procedures, 7 hours of DNA sequencing and 1 hour of data analysis).
